PRESCRIPTION REQUIRED — For veterinary use only. Administer under the strict supervision of a licensed veterinarian. Cyclosporin is a potent immunomodulatory agent; use only after full veterinary diagnosis and at the prescribed dose and frequency. Regular clinical monitoring (renal function, hepatic enzymes, blood pressure, and clinical response assessment) is recommended during long-term therapy. Do not use in animals with active infections, malignancy, or known hypersensitivity to Cyclosporin. Do not administer with grapefruit juice. Keep out of reach of children. Store in a cool, dry place away from direct sunlight; keep bottle tightly capped after use.

Vivaldis Atosporin Cyclosporin Oral Solution for Dogs & Cats (30ml)

Vivaldis Atosporin is a prescription veterinary oral Cyclosporin (Cyclosporine A) solution for dogs and cats. Cyclosporin is a cyclic polypeptide calcineurin inhibitor — the gold-standard oral immunomodulatory agent for the long-term management of canine atopic dermatitis (CAD) and other immune-mediated dermatological conditions. Atosporin oral solution provides Cyclosporin in a convenient liquid format for accurate weight-based dosing in dogs and cats. Administer only under veterinary prescription and supervision with appropriate clinical monitoring. Confirm Cyclosporin concentration (mg/ml) and dosing from the Vivaldis Atosporin product label and veterinary prescription.

Composition

  • Cyclosporin (Cyclosporine A) — confirm mg/ml from product label: Cyclic polypeptide of fungal origin (produced by Tolypocladium inflatum); calcineurin inhibitor class immunomodulatory agent; mechanism of action — Cyclosporin binds to the intracellular receptor protein cyclophilin (also known as peptidyl-prolyl isomerase/PPIA) in T-lymphocytes, forming the Cyclosporin-cyclophilin complex; this complex binds to and inhibits calcineurin (calmodulin-dependent serine-threonine phosphatase), blocking calcineurin-mediated dephosphorylation of the nuclear factor of activated T-cells (NFAT); dephosphorylation of NFAT is required for its nuclear translocation and subsequent transcription of key pro-inflammatory cytokine genes; Cyclosporin inhibition of NFAT nuclear translocation blocks transcription of IL-2 (interleukin-2 — the primary T-cell growth and survival factor), as well as IL-4, IL-5, IL-13, IFN-γ, and TNF-α; suppression of IL-2 prevents T-lymphocyte clonal expansion and activation, and blocks the IL-2-driven amplification loop that sustains chronic skin inflammation in atopic dermatitis; Cyclosporin additionally inhibits mast cell and eosinophil activation, reducing histamine, prostaglandin, and leukotriene release; reduces IgE-mediated skin sensitisation by inhibiting Th2 lymphocyte IL-4 and IL-13 production; inhibits Langerhans cell antigen presentation in skin; collectively these mechanisms reduce both the acute pruritic response and the chronic inflammatory skin changes (lichenification, hyperpigmentation, acanthosis) of atopic dermatitis
  • Oral solution excipients — confirm from Vivaldis Atosporin product label; Cyclosporin oral solutions typically use a lipid-based vehicle (e.g. alcohol, Labrasol, Cremophor EL, or polyoxyl castor oil) to enhance gastrointestinal absorption of the highly lipophilic Cyclosporin molecule; absorption may vary with food — confirm fed vs fasted administration from label

Therapeutic Class

  • Prescription veterinary calcineurin inhibitor — oral immunomodulatory solution for dogs and cats — T-lymphocyte IL-2/cytokine suppression — atopic dermatitis, immune-mediated dermatological conditions

Indications

  • Canine atopic dermatitis (CAD) — Cyclosporin is approved and widely used as a first-line systemic immunomodulatory therapy for CAD; reduces pruritus, skin inflammation, and CADESI (Canine Atopic Dermatitis Extent and Severity Index) scores; onset of full clinical effect typically 4–6 weeks; once controlled, dose may be tapered to alternate-day or twice-weekly administration for long-term maintenance
  • Feline atopic skin syndrome (FASS) — Cyclosporin is used off-label for feline atopy, eosinophilic granuloma complex, and other immune-mediated feline skin diseases; confirm feline dosing from product label and veterinarian
  • Perianal fistulae (anal furunculosis) in dogs — Cyclosporin is the treatment of choice for perianal fistulae in dogs (particularly German Shepherd Dogs), often combined with ketoconazole to inhibit hepatic CYP3A4 metabolism of Cyclosporin and reduce required dose
  • Immune-mediated keratoconjunctivitis sicca (KCS/dry eye) — systemic Cyclosporin as adjunct to topical Cyclosporin ophthalmic preparations in refractory immune-mediated KCS
  • Other immune-mediated dermatological conditions — sebaceous adenitis, sterile nodular panniculitis, and other immune-mediated skin diseases as directed by a veterinary dermatologist
  • Corticosteroid-sparing maintenance — Cyclosporin allows reduction or elimination of long-term corticosteroid use in dogs and cats with chronic atopic and immune-mediated skin disease, reducing steroid-associated adverse effects (PU/PD, polyphagia, iatrogenic Cushing's syndrome)

Dosage

  • Administer orally as directed by a licensed veterinarian based on species, body weight, and clinical condition
  • Standard canine CAD dose: 5mg/kg once daily; confirm from Vivaldis Atosporin product label and veterinary prescription
  • Once clinical remission is achieved (typically 4–6 weeks), dose frequency may be tapered to alternate-day or twice-weekly administration for long-term maintenance
  • Administer consistently in relation to meals as directed on label — confirm fed vs fasted administration from product label
  • Do not administer with grapefruit juice — grapefruit juice inhibits intestinal CYP3A4 and significantly increases Cyclosporin bioavailability and plasma concentration

Drug Interactions (confirm with veterinarian)

  • Ketoconazole, itraconazole — inhibit hepatic CYP3A4 metabolism of Cyclosporin, increasing Cyclosporin blood levels; veterinarians may use this interaction intentionally (ketoconazole combination) to reduce the effective Cyclosporin dose required; requires careful dosing adjustment
  • Fluoroquinolones (enrofloxacin), macrolides (erythromycin) — may increase Cyclosporin plasma levels
  • Phenobarbital, rifampicin — induce CYP3A4 and may reduce Cyclosporin plasma levels below therapeutic range
  • NSAIDs, aminoglycoside antibiotics, amphotericin B — potential for additive nephrotoxicity; use with caution under veterinary monitoring

Safety Information

  • Prescription veterinary medicine — use under strict veterinary supervision with appropriate baseline and monitoring investigations (renal function — BUN, creatinine; hepatic enzymes; blood pressure; clinical response assessment)
  • Do not use in animals with active systemic bacterial, viral, or fungal infections — Cyclosporin's immunosuppressive effects may allow infection exacerbation or dissemination
  • Do not use in animals with known or suspected malignancy — immunosuppression may promote tumour progression
  • Cyclosporin is not recommended during pregnancy or lactation — confirm from label and veterinarian
  • Common adverse effects: gastrointestinal (vomiting, diarrhoea, anorexia, gingival hyperplasia); rare but serious: nephrotoxicity (uncommon at standard dermatological doses), papillomavirus-associated papilloma (warts) during long-term use, opportunistic infections; monitor and report to veterinarian
  • Not for human use — keep out of reach of children
  • Store in a cool, dry place away from direct sunlight; keep bottle tightly capped after use
