Vivaldis Vetecto Spot On is a topical ectoparasiticide for dogs containing Fluralaner — an isoxazoline-class systemic insecticide and acaricide. Unlike conventional topical spot-ons that kill ectoparasites on contact with the skin surface, Fluralaner is absorbed dermally and distributes systemically via the bloodstream, killing ticks and fleas when they bite the treated dog. A single monthly spot-on application provides sustained protection against tick and flea infestations for up to 30 days. The spot-on format provides a no-mess, no-chew alternative to oral isoxazoline tablets for dogs where oral administration is challenging.

Mechanism of Action

  • GABA-gated and glutamate-gated chloride channel antagonism (invertebrate-selective): Fluralaner is an isoxazoline compound that potently antagonises GABA (gamma-aminobutyric acid)-gated chloride channels (GABA−Cl− channels) and glutamate-gated chloride channels (GluCl channels) in invertebrate nervous systems; in insects (fleas) and arachnids (ticks), GABAergic interneurons and motor neurons are tonically regulated by GABA-gated Cl− influx (membrane hyperpolarisation — inhibitory neurotransmission); Fluralaner blocks GABA-gated Cl− influx at insect/arachnid GABA receptor subtypes — specifically binding at the picrotoxin site within the chloride channel pore (RDL-type subunit in insects); channel blockade prevents Cl− influx → sustained membrane depolarisation of invertebrate motor neurons → uncontrolled neuronal excitation → tonic muscle spasms, uncoordinated movement, paralysis, and death of the ectoparasite; Fluralaner concurrently antagonises invertebrate GluCl channels (expressed in invertebrate neuromuscular junctions — absent in vertebrates), compounding ectoparasiticide activity; selectivity for invertebrate vs. mammalian GABA-A receptors (mammalian GABA-A receptors have distinct subunit composition — primarily α1β2γ heteromers with benzodiazepine-sensitive γ-subunits and low isoxazoline binding affinity) provides the safety margin in dogs; CNS penetration in dogs is additionally limited by P-glycoprotein (MDR1/ABCB1) blood-brain barrier efflux — MDR1-deficient breeds (Collies, Australian Shepherds, Shelties, Border Collies, Whippets) may have reduced CNS Fluralaner efflux; at therapeutic spot-on doses this is generally well-tolerated but veterinary guidance is recommended for MDR1-deficient breeds
  • Systemic distribution following dermal absorption: Following spot-on application at the base of the skull/back of neck, Fluralaner is absorbed transdermally into the dermal vasculature and distributed systemically via plasma; Fluralaner is highly lipophilic and extensively plasma protein-bound; it achieves plasma concentrations sufficient to kill Ixodes, Rhipicephalus, Haemaphysalis, and Amblyomma ticks within 12–48 hours of attachment and Ctenocephalides fleas within 8–12 hours; the long plasma half-life of Fluralaner in dogs (≈15 days) maintains ectoparasiticidal plasma concentrations above the killing threshold for the full 30-day inter-application interval from a single spot-on application; unlike surface-contact topicals (e.g. Fipronil), systemic distribution means ectoparasite kill does not depend on the ectoparasite physically contacting the treated skin surface — it requires the ectoparasite to feed on the dog’s blood
  • Spectrum of activity — ticks: Ixodes ricinus and Ixodes scapularis (Lyme disease, Anaplasmosis, Babesiosis vectors), Rhipicephalus sanguineus (Ehrlichiosis, Babesia canis vectors — primary tick species in India), Amblyomma americanum (Ehrlichiosis, RMSF vector), Haemaphysalis spp. (Kyasanur Forest Disease vector — India-relevant); adult and nymphal ticks killed within 12–48 hours of attachment (typically before sufficient feeding time for pathogen transmission for most tick-borne diseases)
  • Spectrum of activity — fleas: Ctenocephalides felis (cat flea — primary flea species infesting dogs in India) and Ctenocephalides canis (dog flea); adult fleas killed within 8–12 hours of attachment and feeding

Indications

  • Treatment and prevention of tick infestations (Ixodes, Rhipicephalus, Amblyomma, Haemaphysalis spp.) in dogs
  • Treatment and prevention of flea infestations (Ctenocephalides felis, C. canis) in dogs
  • Prevention of tick-borne diseases (Ehrlichiosis, Babesiosis, Lyme disease, Anaplasmosis) by rapid tick kill before pathogen transmission
  • Monthly ectoparasite prevention in dogs in tick and flea-endemic regions (including India)

Dosage & Application

  • Topical spot-on: apply entire contents of the weight-appropriate pipette directly to skin at the base of the skull / back of neck
  • Part the fur to ensure direct skin contact; do not apply to broken, inflamed, or irritated skin
  • Apply once monthly; do not bathe or allow swimming for 48 hours post-application to maximise dermal absorption
  • Confirm weight-appropriate pipette size with a veterinarian before first use

Contraindications & Safety

  • Do not use in cats — formulated for dogs only
  • Do not use in puppies under 8 weeks of age or under 2kg body weight (confirm label minimum age/weight)
  • Use with caution in dogs with a history of seizures or neurological disorders
  • Use with veterinary caution in MDR1/ABCB1-deficient breeds (Collies, Australian Shepherds, Shelties, Border Collies, Whippets)
  • Avoid contact with eyes and mucous membranes during application
  • Keep treated dogs away from water bodies — Fluralaner (isoxazoline) has ecotoxicological activity in aquatic invertebrates; do not allow treated dogs to swim in natural water bodies for 48 hours post-application
  • Most common adverse effects: mild transient skin reactions at the application site; occasional GI signs (vomiting, diarrhoea) if product is accidentally ingested by licking
  • Store in a cool, dry place away from heat, flame, and direct sunlight
