PRESCRIPTION VETERINARY MEDICINE — For veterinary use only. Not for human use. MUST BE ADMINISTERED BY OR UNDER THE DIRECT SUPERVISION OF A LICENSED VETERINARIAN — subcutaneous injection requires proper veterinary technique. CRITICAL: CONTRAINDICATED IN MDR1/ABCB1 MUTATION-POSITIVE HERDING BREEDS (Rough/Smooth Collie, Shetland Sheepdog — Sheltie, Australian Shepherd, Border Collie) — P-glycoprotein deficiency allows Ivermectin to cross the blood-brain barrier causing potentially fatal CNS neurotoxicity (mydriasis, ataxia, coma, respiratory depression). CATS ARE HIGHLY SENSITIVE TO IVERMECTIN — injectable Ivermectin must be dosed with extreme care in cats; the therapeutic index is very narrow; use under strict veterinary supervision only. Heartworm endemic areas: Dirofilaria antigen test mandatory before use in non-protected dogs — administering Ivermectin SC to dogs with high microfilarial burdens can cause anaphylactoid shock. Do not use in puppies or kittens under 6 weeks. Keep out of reach of children. Store in a cool, dry place below 25°C. Protect from light. Do not freeze.

Alembic Mectin Pet Injection for Dogs & Cats | Ivermectin 1% w/v | Broad-Spectrum Antiparasitic | Mange, GI Worms, Lungworm & Ectoparasites | 1 Vial x 1ml

Alembic Mectin Pet Injection contains Ivermectin 1% w/v (10mg/ml) as a sterile subcutaneous injectable solution for veterinary use. Ivermectin is a semisynthetic macrocyclic lactone endectocide (active against both endo- and ectoparasites) derived from Avermectin B1 (Streptomyces avermitilis fermentation) that selectively targets glutamate-gated chloride ion channels (GluCl) present exclusively in invertebrate nerve and muscle cells, causing irreversible paralysis and death of susceptible parasites. The subcutaneous (S/C) injectable route provides rapid systemic absorption with high bioavailability and allows precise weight-based dosing of the 1% formulation. Administered strictly as directed by a licensed veterinarian.

Active Ingredient & Mechanism of Action

  • Ivermectin 1% w/v (10mg/ml) — Macrocyclic lactone endectocide (GluCl positive allosteric modulator/irreversible agonist): Ivermectin is a semisynthetic mixture of Avermectin B1a (~80%) and B1b (~20%) — 22,23-dihydro derivatives of the Avermectin B1 macrocyclic lactone natural products from Streptomyces avermitilis fermentation; injectable Ivermectin 1% (10mg/ml in propylene glycol or glycerol formal vehicle) administered subcutaneously is rapidly absorbed from the SC injection site and distributed to all body tissues including the skin (high concentrations in skin and subcutaneous fat — relevant for ectoparasite kill), GI tract wall (relevant for GI nematode kill), and pulmonary vasculature (relevant for microfilarial kill); mechanism: Ivermectin selectively binds and positively allosterically modulates glutamate-gated chloride ion channels (GluCl — Cys-loop receptor family, invertebrate-specific, encoded by invertebrate avr-14/avr-15 and glc-1 genes in nematodes; absent from vertebrate CNS where there are no GluCl channel subunit genes) in nematode pharyngeal and body wall muscle cells and arthropod nerve and muscle cells; Ivermectin binds at the transmembrane interface between GluCl α-subunits → sustained, irreversible (at therapeutic concentrations) opening of the GluCl Cl− channel pore → massive Cl− influx → membrane hyperpolarisation → inhibition of action potential generation → pharyngeal pump paralysis (feeding arrest and starvation) and body wall flaccid muscle paralysis → death of adult worms, larvae, and ectoparasites; Ivermectin also potentiates GABA-gated Cl− channel (GABA-Cl/GABA-A-like receptor) activity at invertebrate neuromuscular junctions, augmenting GABAergic inhibitory tone → additional inhibitory paralytic effect; vertebrate CNS safety: (a) GluCl channels absent from vertebrate CNS; (b) Ivermectin has very low affinity for vertebrate GABA-A receptors at therapeutic concentrations; (c) the blood-brain barrier (BBB) P-glycoprotein (Pgp — ABCB1 encoded by MDR1 gene — expressed on the luminal surface of brain capillary endothelial cells) actively effluxes Ivermectin from the CNS back into the blood, maintaining CNS Ivermectin concentrations well below the threshold for GABA-A receptor-mediated CNS toxicity; MDR1/ABCB1 nt230del4 frameshift mutation herding breed neurotoxicity: see safety warning above; SC vs oral pharmacokinetics: subcutaneous Ivermectin 1% injection achieves higher peak plasma concentrations (Cmax) and longer duration of exposure (AUC) than equivalent oral doses in dogs and cats due to the depot effect of the SC injection site (slow absorption from the propylene glycol vehicle) and avoidance of first-pass hepatic metabolism; this pharmacokinetic advantage allows the injectable formulation to reach therapeutic concentrations against tissue-migrating larvae (e.g., somatic Toxocara canis larvae) and ectoparasites in skin that may be inadequately reached by lower oral bioavailability; dosage: standard antiparasitic dose 200–400 mcg/kg (0.2–0.4mg/kg) body weight SC once; equivalent to 0.02–0.04ml/kg of the 1% (10mg/ml) formulation; 1ml vial = 10mg Ivermectin; for a 10kg dog at 200mcg/kg dose = 2mg Ivermectin = 0.2ml of 1% solution

Parasite Spectrum

  • Gastrointestinal nematodes: Toxocara canis, Toxocara cati (roundworms — adults and L3/L4 larvae), Toxascaris leonina; Ancylostoma caninum, Ancylostoma tubaeforme (hookworms — adult stages); Trichuris vulpis (whipworm — partial); Strongyloides stercoralis
  • Lungworms: Oslerus osleri (Filaroides osleri), Aelurostrongylus abstrusus (feline lungworm — particularly in cats), Crenosoma vulpis
  • Heartworm (Dirofilaria immitis): Microfilaricidal (kills circulating microfilariae); L3/L4 larval preventative; NOT adulticide — adult D. immitis in pulmonary arteries require Melarsomine for adulticide treatment
  • Eye worms: Thelazia spp. (spirurid nematodes of the conjunctival sac)
  • Sarcoptic mange (Sarcoptes scabiei var. canis): Highly effective at 400mcg/kg SC; typically 2–3 doses 2 weeks apart
  • Demodectic mange (Demodex canis): Effective at higher daily or weekly doses (off-label; 400–600mcg/kg); confirm from Alembic Mectin Pet label for approved dose regimen
  • Otodectic mange (Otodectes cynotis — ear mites): Effective systemically via SC route
  • Ticks and lice: Contact kill of ticks and lice present on the animal at time of peak Ivermectin skin concentrations; note — Ivermectin SC does not provide the sustained 4-12 week tick and flea preventative efficacy of isoxazoline oral products (Fluralaner, Lotilaner) — it is primarily used therapeutically for parasites already present

Administration

  • Administer strictly as directed by a veterinarian — SC injection only; do not administer intravenously (IV) — rapid IV administration can cause cardiovascular collapse
  • Standard dose: 200–400 mcg/kg body weight SC — calculate volume from 1% (10mg/ml) formulation
  • Administer SC in the loose skin of the neck or behind the shoulder; use a fine-gauge needle (23–25G); aspirate before injection to confirm SC placement
  • In MDR1-tested sensitive breeds: confirm breed MDR1 genotype before administration; if MDR1(-/-) confirmed, do not administer; if MDR1(+/-), use with extreme caution at minimum effective dose under direct veterinary supervision with post-injection monitoring for 8–12 hours
  • Heartworm antigen test mandatory before use in non-protected dogs in heartworm-endemic areas (India — D. immitis is endemic)

Storage & Handling

  • Store below 25°C; protect from light; do not freeze
  • Once opened, use immediately or store as directed; do not use discoloured or cloudy solution
  • Handle with care — accidental human self-injection requires immediate medical attention; Ivermectin is not approved for human parenteral use
  • Keep out of reach of children
