Sucralfate Oral Suspension is a gastroprotective agent formulated to support the healing of gastric and duodenal ulcers. In an acidic environment, sucralfate reacts with hydrochloric acid in the stomach to form a cross-linked, viscous, paste-like substance. This material acts as an effective acid buffer, providing protection for up to 6–8 hours after a single dose.

The formulation adheres selectively to proteins present on the ulcer surface, such as albumin and fibrinogen, forming stable, insoluble complexes. These complexes create a protective barrier over the ulcer site, shielding it from further irritation caused by gastric acid, pepsin, and bile salts.

Additionally, sucralfate helps prevent the back diffusion of hydrogen ions and adsorbs harmful agents such as pepsin and bile acids. It also promotes the production of bicarbonate, contributing to its acid-neutralizing and cytoprotective effects.

Recent evidence suggests that sucralfate may further enhance mucosal defense by stimulating the production of prostaglandin E2, epidermal growth factors (EGF), basic fibroblast growth factor (bFGF), and gastric mucus. These combined actions support mucosal repair and accelerate the healing process of gastrointestinal lesions.
