Composition & Mechanism of Action

• Clindamycin — lincosamide-class antibiotic. Clindamycin binds to the 50S subunit of the bacterial ribosome, at or near the peptidyl transferase centre, and blocks the formation of peptide bonds between amino acids. Protein synthesis halts, and the bacterium can neither grow nor repair itself. It is generally bacteriostatic, though bactericidal against highly susceptible organisms at higher concentrations. Because the 50S ribosomal subunit is specific to bacteria and structurally different from the mammalian ribosome, host cells are unaffected. Clindamycin is highly lipid-soluble, which lets it penetrate bone, abscess walls, and soft tissue where many other antibiotics cannot reach adequate concentrations. Its spectrum covers Gram-positive aerobes including Staphylococcus and Streptococcus species, and a broad range of anaerobes including Bacteroides, Fusobacterium, Clostridium, and Peptostreptococcus.
