Enterococcus faecium — Probiotic Mechanisms: E. faecium is one of the most extensively studied and validated veterinary probiotic strains (SF68 strain widely used in companion animals); mechanisms: (1) competitive exclusion — E. faecium colonises intestinal epithelial adhesion sites, competitively excluding enteric pathogens (E. coli, Salmonella, Clostridium perfringens, Campylobacter) from binding and colonising the mucosa; (2) lactic acid and bacteriocin production — E. faecium produces L-lactic acid (reducing luminal pH, inhibiting acid-sensitive pathogens) and bacteriocins (enterocins — antimicrobial peptides targeting Gram-positive pathogens including Listeria, Staphylococcus, and Clostridium); (3) tight junction reinforcement — E. faecium upregulates expression of tight junction proteins (occludin, claudin-1, ZO-1) at intestinal epithelial cell junctions, reducing intestinal permeability (‘leaky gut’) and preventing bacterial translocation; (4) immune modulation — E. faecium interacts with intestinal dendritic cells and Peyer’s patch macrophages via TLR-2 and NOD2 pattern recognition receptors, promoting tolerogenic immune responses (Treg/IL-10/IL-12 balance) and mucosal IgA secretion; (5) restoration of post-antibiotic dysbiosis — E. faecium is naturally resistant to several antibiotics and survives antibiotic treatment courses, providing a seeding population for microbiome restoration during and after antibiotic therapy

FOS — Fructooligosaccharides (Prebiotic): FOS are short-chain β-fructose polymers (degree of polymerisation 2–8); selectively fermented by Bifidobacterium longum, B. animalis, Lactobacillus acidophilus, and E. faecium in the large intestine; fermentation produces short-chain fatty acids (SCFA) — principally butyrate, propionate, and acetate; butyrate is the primary energy substrate for colonocytes (large intestinal epithelial cells), supporting colonocyte proliferation, mucosal barrier repair, and anti-inflammatory effects via inhibition of NF-κB and HDAC (histone deacetylase) in colonocytes; FOS increase Bifidobacterium and Lactobacillus relative abundance while reducing Clostridium, Enterobacteriaceae, and Bacteroides populations; FOS also increase faecal water content and stool bulk, improving stool consistency in both constipated and diarrhoeic dogs
